Direct Connection Sync Two Computers

I want to use direct connection to sync my laptop and desktop computers. Does DevonThink have to be running on both computers for this to work?

I must admit a little paranoia about having DevonThink open on two machines during syncing.

Yes but all other options (Dropbox, WebDAV, local sync stores) don’t require this.
