Thought this was a bug on 2.10.0 but remains on 2.10.1
iCloud is turned on on both of my computers. Both have the same issue.
I’m probably missing something.
Is DEVONthink enabled in System Preferences > iCloud > iCloud Drive > Options?

DevonThink Pro Office 2.10.1
Does it work after downloading it again from our website and replacing the installed version with the downloaded one? Otherwise a screenshot of System Preferences > iCloud > iCloud Drive > Options might be useful.

Which version of macOS do you use?
High Sierra on one, Sierra on the other. Both don’t work.
Is there any third-party software installed on both computers that might patch the system, e.g. stuff like VM (e.g. Parallels) or other haxies? Does it work using a new clean user account (see System Preferences > Users & Groups to create one)?
