No, this isn’t possible (yet).

Not that I’m aware of. The best solution is to pass-word protect the database. Once you open that database along with all your other databases, you’ll likely not notice it’s a different database.
Password protecting Groups (they are not macOS folders) is another matter and I can easily imagine the issues that prevent that from being a viable option for DEVONthink.
You could experiment with using macOS features (and other 3rd party apps) to password protect macOS folders. I’m not sure what would happen if you then tried to index them into DEVONthink, but perhaps that not really needed. If data is that sensitive to protect beyond full disk encryption of the Mac, then best perhaps not to spread it around too much. Just a thought.
